Admissions to 12th are not controlled by state but eligibility is decided by the university. For B.Sc in biotechnology, bio is desirable but not mandatory in 12th.

Entrance exam required is the GRE and TOEFL for Master's degree courses in USA. Taking the GRE and TOEFL will not guarantee a place at Harvard, however, one of the top ranking universities and highly competitive. They seek all rounders hence prepare accordingly.

You have to understand the difference between an M.Sc or BE/B.Tech and decide accordingly. I also fail to understand why you want to pursue a BE or B.Tech now that you already hold a B.Sc? Secondly, I don't see any reason why you won't be admitted to an MS programme in biotechnology if you meet admission requirements. Check university websites for the admission criterion, take your IELTS or TOEFL. GRE would be necessary to study in US.
