Generally similar stream will be offered for B.Tech for Diploma students as studied in Diploma. However, B.Tech Biotechnology basically deals with basic engineering concepts which are applied in chemical engineering and applied Biology. B.Tech Biotechnology Syllabus also deals with methods for environmental conservation, medicines, health along with technology. So changing the stream in B.Tech course is not recommendable.

As you will find a good number of colleges to pursue B.Sc. In biotechnology so, to choose one of the best to suit your requirement is little tedious. I suggest you for IMSUC Ghaziabad for hons. As you might come across with few good names so, compare them all to take decision for the best suitable one for you.

Yes, you can pursue a course like MCA irrespective of your graduation background. I have seen people with Biology honors in Graduation have moved to the software domain by doing MCA degree. So, you can also do it but do the MCA course from a well-reputed institute at the state level or a national level.
