It is interesting to read brochure of IISc. It says, candidates will be selected on the basis of merit list that will be based on any one of the following national level tests. This includes JEE Mains and JEE Advanced.

Yes, you can get admission in IISc without appearing for JEE Mains or Advanced, but you will have to appear for some other all India level exam. Why don’t you want to appear in JEE Mains? You can still apply.
However, IISc is expected to open online application forms for admission in the 2nd week of February, you have to wait till then, for details. But remember that admission is held on the basis of your score in any of the entrance exams you appear for.

You have to prepare for IIT JAM Entrance test to pursue M.Sc. from IISC. The eligibility required for IISC is general and OBC candidates:
First class marks or min. 60% aggregate marks, without rounding off, in qualifying degree.
SC/ST/PWD Candidates:
Second class or min. 50% aggregate marks without rounding off, in qualifying degree.
The candidates who have either appeared or are due to appear in the final examination of their qualifying degree in 2017 are also eligible to appear in the test.
